Lumi-Phos Plus Description
Lumi-Phos® Plus is a ready-to-use chemiluminescent substrate formulated for the sensitive detection of alkaline phosphatase (AP) conjugates on solid supports. Containing Lumigen PPD and a patented enhancer in 2-amino-2-methyl-1-propanol buffer, the reagent is optimized for nucleic acid detection applications including Southern and Northern blotting, dot blots, colony and plaque screening, and cDNA array analysis.
The substrate produces bright blue chemiluminescence with maximum emission at 470 nm. It is suitable for direct detection of AP-labeled nucleic acids or indirect detection of hapten-labeled probes using AP-conjugated anti-hapten antibodies. Lumi-Phos Plus delivers enhanced performance characterized by low background signal, fast signal ramp-up, and high-intensity light output.
In alkaline conditions, the chemiluminescent signal increases over time and reaches a stable plateau, supporting flexible exposure windows for film or digital imaging systems. At lower enzyme concentrations, the sustained luminescence remains stable for extended periods, enabling reliable detection of low-abundance targets. Compared to Lumi-Phos 530, Lumi-Phos Plus provides an improved signal-to-background ratio due to its reduced intrinsic reagent background.
The formulation is designed to provide consistent, reproducible performance in homogeneous aqueous solutions and on nylon or nitrocellulose membranes. Its stability and long shelf life support routine use in research and diagnostic assay development workflows.
